ABSTRACT

ObjectiveTo discuss the diagnostic methods of global developmental delay caused by 10q24.3 heterozygous loss. MethodsA retrospective analysis was conducted on the clinical data of one child with global developmental delay, and the results of low depth whole-genome copy number variation sequencing (CNVseq) and family whole exome sequencing (WES) of the child and his parents. ResultsThe patient was a 10-month-old male with developmental retardation in four areas, with some special features (ocular hypertelorism, strabismus, flat nose bridge, protruding forehead, cleft palate, high palatal arch, etc.) and hypotonia of limbs. The CNVseq and WES test showed that the patient had new 10q24.3 heterozygosis loss. Because this region contains the gene SUFU associated with basal cell nevus syndrome and the gene CNNM2 associated with hypomagnesemia, seizures, and mental retardation, and the gene TRIM8 associated of Focal segmental glomerulosclerosis with neurodevelopmental syndrome, we speculated that the cause of the disease in the child was highly related to the heterozygosity deletion of SUFU gene and CNNM2 gene and TRIM8 gene. ConclusionGenetic testing should be improved as soon as possible for children with global developmental delay and special facial manifestations, so as to make clear diagnosis and to judge prognosis.

ABSTRACT

@#A pre-driving assessment usually involves standardised and non-standardised tests to examine the skills for safe driving of people with disabilities. However, it is still unknown to what extent the pre-driving assessments are being practised among occupational therapists in Malaysia. Therefore, this study aims to investigate the association of challenges, attitudes, and skills of occupational therapists towards practising driving rehabilitation and explore the most used predriving assessment. A cross-sectional study had been conducted from July to September 2020. The convenient sampling method was used to recruit experienced occupational therapists in driving rehabilitation. A total of 53 therapists had completed an online survey. According to the results, the bivariate correlation between skills and challenges was positive and strong (rѕ = 0.680, p < 0.05). A multivariate analysis of variance (MANOVA) indicated that workplace variables had a significant effect on the combined dependent variables of challenges, attitudes, and skills, F(3,49) = 4.521 p = 0.007, partial Ƞ² = 0.217. This study found that the most used standardised assessment was the range of motion (n = 39, 73.6 %) and the right and left discrimination (n = 40, 75.5 %). The findings of this study identified that occupational therapists challenges and attitudes are significantly associated with the skills to practice driving rehabilitation. Malaysian occupational therapists working in the government sector reported better skills than those working in the private sector. Furthermore, numerous types of pre-driving assessments were used and vary from setting to setting. This study hopes to benefit occupational therapists and related rehabilitation practitioners towards a holistic understanding of Malaysia's driving rehabilitation practices.

ABSTRACT

The present study explored the drying effect of new spiral vibration drying technology on Chinese medicinal pills with Liuwei Dihuang Pills, Zhuanggu Guanjie Pills, and Muxiang Shunqi Pills as model drugs. With the drying uniformity, drying time, energy consumption, pill split, dissolution time, and change of index components as evaluation indicators, the drying effect of spiral vibration drying technology on model drugs was evaluated and compared with traditional drying methods, such as hot air drying and vacuum drying in the oven. The dynamic changes of moisture in Liuwei Dihuang Pills with different drying time were investigated. Compared with the traditional drying methods in the oven(hot air drying and vacuum drying) at 80 ℃, the spiral vibration drying only took 80 min, shortened by 80%, with 10%-13% energy consumed. The results showed that the moisture of Liuwei Dihuang Pills was negatively related to the drying time. By virtue of multi-layer countercurrent drying and super resonant fluidization techniques, the new spiral vibration drying technology can significantly improve the drying quality of Chinese medicinal pills, improve the drying efficiency, and enhance the manufacturing capacity of Chinese medicinal pills. This study is expected to provide references for the innovation and development of new drying technology of Chinese medicinal pills.

ABSTRACT

Drying is one of the most common unit operations in the production of traditional Chinese medicine. The drying process of traditional Chinese medicine materials is accompanied by the dynamic reduction of water content. As a key index to determine the end of the drying process, the moisture content of materials plays an important role in improving drying efficiency and saving energy. Recently, the drying process of traditional Chinese medicine is mostly monitored by offline detection, and there are few reports of online moisture detection applications. In this paper, the principle and current application of online inspection technology for the material drying process in different fields were introduced. The significance of online detection technology in drying of traditional Chinese medicine was also analyzed. Meanwhile, the application prospect of online detection technology in the field of drying of traditional Chinese medicine was predicted. In response to urgent transformation and upgrading of the traditional Chinese medicine manufacturing industry, the application of online moisture detection technology is expected to be a key breakthrough in the intelligent upgrading of traditional Chinese medicine drying technology and equipment.

ABSTRACT

Purpose@#This study aimed to investigate the potential systemic antitumor effects of stereotactic ablativeradiotherapy (SABR) and apatinib (a novel vascular endothelial growth factor receptor2 inhibitor) via reversing the immunosuppressive tumor microenvironment for lung carcinoma. @*Materials and Methods@#Lewis lung cancer cells were injected into C57BL/6 mice in the left hindlimb (primary tumor;irradiated) and in the right flank (secondary tumor; nonirradiated). When both tumors grewto the touchable size, mice were randomly divided into eight treatment groups. These groupsreceived normal saline or three distinct doses of apatinib (50 mg/kg, 150 mg/kg, and 200mg/kg) daily for 7 days, in combination with a single dose of 15 Gy radiotherapy or not tothe primary tumor. The further tumor growth/regression of mice were followed andobserved. @*Results@#For the single 15 Gy modality, tumor growth delay could only be observed at the primarytumor. When combining SABR and apatinib 200 mg/kg, significant retardation of both primaryand secondary tumor growth could be observed, indicated an abscopal effect wasinduced. Mechanism analysis suggested that programmed death-ligand 1 expressionincreased with SABR was counteract by additional apatinib therapy. Furthermore, whenapatinib was combined with SABR, the composition of immune cells could be changed.More importantly, this two-pronged approach evoked tumor antigen–specific immune responsesand the mice were resistant to another tumor rechallenge, finally, long-term survivalwas improved. @*Conclusion@#Our results suggested that the tumor microenvironment could be managed with apatinib, which was effective in eliciting an abscopal effect induced by SABR.

ABSTRACT

Background: Baicalein is an active compound extracted from the roots of Scutellaria baicalensis georgi, which is widely and traditionally used in the anticancer therapy. Notch signaling pathway is usually abnormally activated in kinds of human cancers. The aim of the present study is to investigate the antitumor effects of baicalein in human cervical cancer and explore whether baicalein treatment affects notch signaling pathway in human cervical cancers. Materials and Methods: Cervical cancer cells were treated with increasing concentrations of baicalein for 24, 48, and 72 h, respectively. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to determine cell viability of cervical cancer cells. The apoptosis rate was determined by FACS assay. Furthermore, the molecular mechanism was investigated. The expression levels of Notch 1, Notch 2, Notch 3, hairy enhancer of split-1 (Hes-1), and Hes-5 were determined by western blotting analysis. Results: MTT assay results revealed that baicalein inhibited cell proliferation of HeLa cells and SiHa cells in a time- and dose-dependent manner. The data from FACS assay demonstrated that baicalein-induced cell apoptosis of cervical cancer cells at the final concentration of 100 μM for 24 h. Furthermore, baicalein treatment downregulated Notch 1/Hes-1, Hes-5 signaling pathway, and there was no obvious change on the expression of Notch 2 and Notch 3. Conclusion: Baicalein inhibited the proliferation of human cervical cancer cells via Notch 1/Hes signaling Pathway. The study would provide some new clues in the clinical therapy of human cervical cancers

ABSTRACT

Objective: Taking single Liuwei Dihuangwan(LDW) as the research object, the moisture content change and volumetric shrinkage characteristics in its drying process were investigated, which provided the theoretical basis for improving the drying efficiency of the pills and reducing the quality problems of the pattern pills, crusts and crack pills. Method: The drying characteristics of LDW at drying temperature of 50,75,100,125℃ were studied by constant temperature hot air drying and vacuum drying.Based on sphere model of Fick's second law and Arrhenius equation, the effective diffusion coefficient and activation energy of the drying process were obtained. The volumetric shrinkage characteristics of the pills during the drying process were studied by the projected area method. The Weibull function was used to fit the drying dynamics curve of single LDW. Result: Hot air drying and vacuum drying of LDW both belonged to the decreasing drying processes, and the time required to achieve the same moisture content in vacuum drying was shorter than that in hot air drying. The moisture ratio in the drying process of single LDW obeyed the Weibull function distribution(R2=0.994 5-0.999 7),the scale parameter(α) decreased with the increase of temperature, and drying temperature had significant influence on the shape parameter (β).The effective diffusion coefficients of hot air drying and vacuum drying were 2.626×10-3-7.823×10-2,3.782×10-3-9.042×10-2 m2·s-1, their activation energy were 47.18,42.69 kJ·mol-1,respectively.The volume ratio of hot air drying and vacuum drying of LDW ranged from 0.638 to 0.741 and 0.607 to 0.689,respectively. Conclusion: Weibull function can be adopted to predict the drying and dehydration law of LDW.Under the condition of low temperature drying,slow-down of shrinkage rate of the pills is helpful to prevent the formation of splitting pills, this study provides theoretical and technical basis for dying of LDW.

ABSTRACT

Dry granulation technology is a great innovation in granulation technology, which saves many intermediate links and reduces many intermediate costs. It is closely related to the characteristics of materials, dry granulation equipment and process. Dry granulation technology is a systematic engineering science covering many technical fields. The process of dry granulation involves complex mathematical model mechanisms of temperature field, pressure field and velocity field, closely related to the characteristics of materials and drying equipment. However, due to the late start of research on dry granulation technology of traditional Chinese medicine, basic research is still weak. The research on dry granulation technology has achieved great results in the fields of food, chemical industry, agriculture and forestry, showing great reference significance. The advantage of dry granulation of traditional Chinese medicine is that it can be directly granulated by adding an appropriate amount of auxiliary materials in the extract powder of traditional Chinese medicine, without the need of wetting, mixing, drying and other processes. The process is simple and can effectively guarantee the quality of traditional Chinese medicine. The granules obtained by the dry granulation technique are important intermediates for preparing the solid preparations of traditional Chinese medicines, which would directly affect the subsequent molding process and the quality of the preparation products. Therefore, based on the characteristics of dry granulation method in traditional Chinese medicine and by referring to the advanced research results of dry granulation technology in other fields,we would discuss the research ideas of dry granulation in traditional Chinese medicine in terms of the mechanism of dry granulation equipment,technology,on-line detection technology and mathematical model of dry granulation process, hoping to provide reference for the research of dry granulation method in traditional Chinese medicine.

ABSTRACT

STUDY DESIGN: In Vitro cell culture study. PURPOSE: This study aims to investigate the impact of transforming growth factor-beta1 (TGF-β1) and lovastatin on differentiating human mesenchymal stem cells (MSCs) toward nucleus pulposus (NP)-like phenotype. OVERVIEW OF LITERATURE: MSCs offer a cell source to the cell-based therapy for intervertebral disc degeneration. TGF-β1 is used to induce MSCs to differentiate into NP-like cells; however, an undesired expression of collagen type I has been reported. Statins reportedly stimulate expression of bone morphogenetic protein-2 (BMP-2) and promote the chondrogenic phenotype to NP cells. However, the effects of statins with or without TGF-β1 on the differentiation of MSCs into NP-like cells remain unclear. METHODS: Human MSCs were treated with TGF-β1 alone, lovastatin alone, and simultaneous or sequential treatment with TGF-β1 and lovastatin. After the proposed stimulation, the total RNA was extracted to assess the expression profile of NP cells-specific genes. Hematoxylin–eosin staining was used for examining the microscopic morphology. Furthermore, we detected the syntheses of S-100 protein, aggrecan, and collagen type II in the extracellular matrix using immunohistochemical staining. RESULTS: Simultaneous or sequential treatment of TGF-β1 and lovastatin could further augment the BMP-2 overexpression compared with lovastatin-alone treatment. However, the mRNA expression of aggrecan and collagen type II was not compatible with the expression level of BMP-2. Immunohistochemical studies revealed compatible production of aggrecan, collagen type II, and S-100 protein in all three groups treated with lovastatin. Cells in groups treated with lovastatin were less populated than that in the group treated with TGF-β1 alone. CONCLUSIONS: This study demonstrates a promising role of lovastatin in inducing human MSCs into NP-like cells. However, further optimization of cell density before lovastatin treatment, treatment duration, and combination with TGF-β1 are warranted to attain better stimulatory effects.

ABSTRACT

PURPOSE: To compare differentially expressed genes (DEGs) mediating osteoarthritis (OA) in knee cartilage and in normal knee cartilage in a rat model of OA and to identify their impact on molecular pathways associated with OA. MATERIALS AND METHODS: A gene expression profile was downloaded from the Gene Expression Omnibus database. Analysis of DEGs was carried out using GEO2R. Enrichment analyses were performed on the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes pathway using the Search Tool for the Retrieval of Interacting Genes database (http://www.string-db.org/). Subsequently, the regulatory interaction network of OA-associated genes was visualized using Cytoscape software (version 3.4.0; www.cytoscape.org). RESULTS: In the gene expression profile GSE103416, a total of 99 DEGs were identified. Among them, 76 DEGs (76.77%) were overexpressed, and the remaining 23 DEGs (23.23%) were underexpressed. GO and pathway enrichment analyses of target genes were performed. Using gene-gene interaction network analysis, relevant core genes, including MET, UBB, GNAI3, and GNA13, were shown to hold a potential relationship with the development of OA in cartilage. Using quantitative real-time PCR, the Gna13/cGMP-PKG signaling pathway was identified as a potential research target for therapy and for further understanding the development of OA. CONCLUSION: The results of the present study provide a comprehensive understanding of the roles of DEGs in knee cartilage in relation to the development of OA.

To review retrospectively six cases of rhino-orbital related endoscopic surgeries aided by Fusion electromagnetic system,to explore the indications and clinical value of image guided technique in endonasal endoscopic surgery.Retrospective research methods were used.In this study, six cases of nasal endoscopic sinus surgery using Fusion electromagnetic system were analyzed, including 1 nasal penetrating foreign body,2 optic nerve decompressions,1 orbital apex hemangioma,1 sieve frontal sinus cyst,1 intraorbital mass biopsy. The preparation time of navigation system, the accuracy of intraoperative positioning and surgical coherence, intraoperative and postoperative complications of surgery were recorded. The average preparation time was(8.13 ± 1.858)min.In the navigation, the sinus ostium, orbital cardboard, skull base, optic nerve, internal carotid artery and other important structures can be accurately located in all patients, while registrations had been accurate within 1 mm.Six patients were successfully operated by image guided technique. There was no intracranial or intraorbital complications due to intraoperation error.Image guided technique allows for a truely microinvasive and accurate rhino-orbital related endoscopic surgeries. It requires less preoperative preparation time, has high surgical navigation accuracy, improves the surgical coherence and safety, and reduces the surgical complicationgs. However,as an auxiliary tool,it can not replace the surgeon's anatomical knowledge, surgical training and clinical experience.

ABSTRACT

In many pathogens infection, especially virus,antibody-dependent enhancement(ADE) can aggravate the infection and lead to severe diseases. In this immunopathological phenomenon,virus-specific antibodies enhance the entry of virus into monocytes, macrophages and granulocytic cells and even the replication of virus through different mechanism. This phenomenon has been reported in numerous pathogens including virus, bacteria and parasite and the mechanisms of ADE vary from different species.Further study of ADE can promote the vaccine research and development to make the most use of vaccine and prevent human body from pathogens, which will be helpful to control the spread of pathogens including Zika virus. In the present review,we review the research progress of ADE mechanism in recent years, including antibodies mediating, receptors mediating, complement mediating, viral proteins mediating and cellular mediating ADE.In addition, dengue virus, human immunodeficiency virus,Coxsackie virus,Ebola virus,Zika virus and other pathogens will be illustrated respectively. This review provides insights on the different mechanism of ADE in different pathogens.
